Antagen的慢病毒滴定XpressCard是一種快速側(cè)流免疫分析，可以快速確認(rèn)包裝上清液中有用的慢病毒滴度*。該測試檢測釋放到包裝上清液中的慢病毒膠囊蛋白作為替代標(biāo)記，以確定病毒產(chǎn)生是否為隨后的感染產(chǎn)生可用的滴度。與需要2-3天的常規(guī)基于再感染的測定相比，該測定僅需10-15分鐘。

*鼓勵用戶將T線強(qiáng)度與其自身感染系統(tǒng)中的功能滴度相關(guān)聯(lián)，因為不同的細(xì)胞類型對慢病毒具有不同的易感性。在上述實施例中，用連續(xù)稀釋的慢病毒包裝的上清液感染HEK293細(xì)胞。72小時后，在熒光顯微鏡下觀察表達(dá)ZsGreen的細(xì)胞。通過含有~2×10 6 IFU / ml的稀釋液（通過轉(zhuǎn)染的HEK293細(xì)胞的流式細(xì)胞術(shù)測量）產(chǎn)生暗紅線。
我們使用已知滴度的純化濃縮慢病毒原種的連續(xù)稀釋來測試我們的XpressCards的靈敏度。結(jié)果顯示檢測限為1×10 5 TU / ml或1×10 4 TU（100μL）。
